Aseptic Transfers and Inoculation Methods

Aseptic transfers and inoculation methods are used to take some bacteria from one culture to another culture. An aseptic transfer technique uses a stock culture (SM) which is a microorganism used to keep the microorganism in question in a good condition so it can be used to transfer to a new medium. Aseptic transfers and inoculations are used to introduce new microorganisms to new conditions suitable for growth (media). The materials used for the aseptic transfer and inoculation are different media, including a slant, deep, broth, and plate/dish. The caps of the media should be a different color so it is easy to differentiate between the three. A Bunsen burner is also used to sterilize the inoculating loop and the inoculating needle to transfer the bacteria into the medium. The Bunsen burner prevents cross contamination. There are some tips to make sure that the transfer of the bacteria goes a little bit easier, such as holding the media tubes in a “V” shape in your non-dominant hand while holding the inoculating tube in your dominant hand. Also, you can prevent contamination of the specimen by holding the tubes facing downward while holding them with your pinky. Also, passing the necks of the tubes through the flame of the Bunsen burner prevents any further contamination. After the bacteria is incubated, the new bacteria will grow in the media we used (slant, deep, broth).The cultures will be transferred to the new media and will prove that the bacteria can be inoculated....
