Amantadine . Carbidopa/ Levodopa

Adverse effects

MODE OF ACTION

Inhibits the reuptake of dopamine by blocking other neurotransmitters such as Monoamine Oxidase , increasing availability of dopamine at the receptor site, or stimulates the production of dopamine depending on which drugs is taken. These drugs will decrease the shakiness of the patient with Parkinson’s disease.

Dry mouth Choreiform movements Dry eyes Increased tremors Dystonic movements of the face

Interactions à

Tricyclic antidepressants: Increased risk of hypertension and dyskinesia

Antacids: Increased effect of levodopa

à

Anticonvulsants: Decreased effect of levodopa

à

Foods high in pyridoxine (vitamin B6) or vitamin B6 preparations reduce the effect of levodopa.

Critical thinking These drugs are common for use in conjunction with anti-psychotic drugs to prevent EPS and tardive dyskinesia.

Contraindications

Levodopa is contraindicated in patients with narrow-angle glaucoma and those receiving MAOI antidepressants. Do not give with meperidine Herbal Considerations

Kava-Kava will decrease the effects of carbidopa/Levodop

May cause an inaccurately false Coombs’ test May cause increased glucose levels May cause elevation in AST or ALT or decrease in BUN Toxicity

Involuntary muscle twitchin Excessive eye blinking

INDICATIONS à Parkinson’s disease à Parkinson-like symptoms (extrapyramidal) as a result of injury, drug therapy, or encephalitis à Restless leg syndrome (RLS) à Viral infections (amantadine)
