INTRODUCTION MYCOLOGY is the study of the eukaryotic fungi, including yeasts, moulds, and mushroom. MEDICAL MYCOLOGY is the study of fungi that produce pathology or disease in humans and animals including their ecology and epidemiology DIAGNOSTIC MYCOLOGY is the study of the general characteristics, pathology & pathogenesis, and laboratory tests & methods of medically important fungi. that aids in their identification and treatment GENERAL CHARACTERISTICS OF FUNGI Fungi constitute an extremely diverse group of organisms and are generally classified as molds or yeasts. Some have been recognized as classic pathogens, whereas others are recognized only as environmental saprobes, living on nonliving material GRAM STAINING REACTION Fungi stains gram positive EUKARYOTIC ORGANISMS Have a true nucleus bounded by a nuclear membrane. Also have endoplasmic reticulum for protein synthesis & mitochondria for energy production o Fungi are Eukaryotic, classified in their own separate kingdom, the Kingdom Fungi o Lack chlorophyll, thus heterotrophic MOTILITY Non motile CELL WALL Definite cell wall that contains complex carbohydrates such as chitin, mannan, glucan, & chitosan Ergosterol present in the cell membrane NOTE! o Grow best at neutral pH o Lack susceptibility to antibacterial agents Table 1 – Fungi Vs. Bacteria Fungi Bacteria Eukaryotic “has Prokaryotic “no Cell Structure
nucleus”
nucleus
Cytoplasmic Components Size Spores basic reproductive unit of fungi Mode of Reproduction
Polysaccharide: Mannan Glucan Chitin Other polysaccharides Sterol (Ergosterol) – components of membrane; site of attacks of antifungal; not selective it will attack the cells and hormones such as testosterone and progesterone causing impotene endoplasmic reticulum (rough), mitochondria >5um for reproduction
Peptidoglycan/ Muramic acid
Sexual rep. Asexual rep
Binary fission
No sterol
no ER, mitochondria...
