Lab of DNA extraction...

Description

AP Biology A DNA Extraction Lab Directions: Begin by watching the video of a DNA Lab Video - Strawberry DNA Extraction and think about how easy it is to do a quick DNA extraction at home. What scientific advances have made this possible? Look over some of the links provided in the Helpful Links (below) and develop a hypothesis and design an experiment that will allow you to perform your own DNA extraction. Materials: • • • • • • • • • • • • • • • •

Measuring cup Measuring spoons Rubbing alcohol Salt water dishwashing detergent (Dawn®) Glass or small bowl Cheesecloth Funnel Tall drinking glass Meat tenderizer DNA source: bananas, peas, broccoli, spinach, or strawberries Resealable plastic sandwich bags Test tube or small glass jar Bamboo skewer Blender

Use the AP Biology Lab Report Rubric to guide your lab report writing. • Write an introduction for your report including background of the experiment, the purpose of the experiment, and an appropriate if / then / because hypothesis. • List the materials needed and write a detailed procedure. Be sure to include a description of the control and experimental group, as well as the independent and dependent variables. • Prepare and record data in a table. You might need to record qualitative data, quantitative data, or both. You will also need to take pictures of your lab and include them in your lab report. • Create graphs based on the data collected. You might need to get a little creative here, but an appropriate graph is required. Write and answer 5 analysis questions based on the background and the purpose of the lab. This is the opportunity for you to think like the teacher and develop some thought provoking questions. • Write a conclusion including the summary of your findings and a discussion of how your hypothesis was supported or refuted.

• Finally, include a listing of the references that were used to develop, execute, and analyze your experiment. Submit your completed lab report. DNA Extraction Strawberry Lab During this lab DNA will be extracted from a strawberry so it can be observed, which is the purpose of this lab. If more dishwashing detergent is used, then more DNA will be extracted because that’s what cause the cell to open and release DNA. The constant in this experiment is the amount of strawberries being used. The independent variable is the amount of dishwashing detergent used. The dependent variable is the amount of DNA extracted. The materials needed for this experiment is rubbing alcohol, measuring cup, measuring spoons, salt, water, dishwashing liquid, small bowl, cheesecloth, funnel, tall drinking glass, strawberries, plastic sandwich bag, a small glass jar, a bamboo skewer, and a blender. First, you make an extraction liquid that includes water, dishwashing detergent and salt. Then, you smash the strawberries in a small bag and add some of the extraction solution. Then the strawberries will be filtered by putting cheesecloth on top of a funnel and filtering it out through that. After that use the strawberry liquid and add the rubbing alcohol and a white stringy substance should arise, which is the DNA....
