ESTIMATION OF TOTAL SUGARS BY
Date: ANTHRONE REAGENT Aim To estimate the amount of total sugars present in the given sample using anthrone reagent. Principle Starch solubilized in perchloric acid is hydrolyzed to glucose in hot acidic medium which undergoes dehydration to form hydroxy methyl furfural. This derivative then condenses with anthrone to give a blue coloured complex and is determined quantitatively by anthrone sulphuric acid procedure.

Materials Boiling water bath Anthrone – sulphuric acid reagent: Dissolve 2g of anthrone in 1 litre of cold 95% con. H2SO4. Store at 4ºC. This solution should be prepared fresh. Procedure 1. Take 1 ml of the unknown solution in a test tubes and place it in a cold water bath. 2. Add 2 ml of a freshly prepared anthrone reagent and vortex. 3. Incubate the tubes in boiling water bath for 7.5 min. 4. After cooling the tubes under running tap water, note the absorbance of these solutions at 630 nm. 5. Prepare a standard graph using 0 – 500 µg glucose as per the procedure described above.
