Lab 1 for microbiology...
Microbiology Lab Safety PRE-LAB QUESTIONS 1. What constitutes personal protective equipment? When should personal protective equipment be worn? Why is personal protective equipment important? Personal protective equipment is gloves, mask, goggles, gown, shoe covering, basically anything that covers you for your protection. PPE should be worn at all times during your experiment. PPE is important because it will protect you from any chemicals or things that may cause harm to you.
2. Why are there increasing levels of biological containment procedures? What level of biological containment will be necessary to complete the experiments in this manual? In this microbiology course, level 1 of biological containments will be used. It summaries how much caution needs to be used when working with the lab materials. It also is a guide to what precautions should be used.
3. What document should you refer to if you have questions regarding the safety or disposal of a chemical? Where can this document be located? This document is the SDS and can be found on the eScience website.
List five safety procedures that should always be adhered to when working with microorganisms. 1. Wash hands before and after experiment 2. No food or drink on your lab table while doing experiment 3. Disinfect your lab area after each lab 4. Goggles, lab coat, and gloves should be used during lab. 5. Long hair should be tied back during lab.

Post-Lab Questions 1. What purpose did plating the yeast serve in this experiment? What purpose did the water serve? The purpose of the yeast is to serve as a similarity to human cells and the water serves as the control group.
2. Which of the two hand treatments grew the most colonies? The hand that was not washed grew the most colonies.
3. What effect does handwashing have on the presence of bacteria? After the experiment, it is clear that washing your hands decreases the colonies growing. Hand washing should kill majority of the bacteria.
4. Were there any colonies present on plate #2? What could account for the presence of these colonies? Yes. Some areas of my hand may not have been washed as thorough as others, causing some bacteria.
