LAB 2: Grading the Reaction Strength OBJECTIVES Following this laboratory exercise, the student will be able to: 1. Grade the agglutination reactions in a uniform manner. 2. Detect hemolysis in a reaction. 3. Record the agglutination reactions and interpretations with accuracy and consistency. PRINCIPLE / DISCUSSION The endpoint for most test methods involving red cells and blood group antibodies is agglutination. Uniform grading of agglutination reactions is important for antibody detection and identification, blood typing, and antibody titers. Recording reactions upon observation and documenting the interpretation correctly are essential for safe transfusion outcomes. Grading and interpretation abbreviations are unique to each lab. Students should become familiar with the importance of following laboratory policies for recordkeeping. Hemolysis is a characteristic reaction to some antibody groups, as well as evidence of other clinical problems. Recognition of hemolysis as a positive reaction of an antigen-antibody complex is important in blood banking. REAGENTS A1 cells PROCEDURE 1. Label twelve test tubes 1 to 12 to correspond to the test samples provided. 2. Add 2 drops of each test serum to its appropriately labeled tube. 3. Add 1 drop of reagent A1 cells to each tube, mix, and centrifuge the tubes (30 seconds). 4. Using the view lamp, gently shake or tilt the tube and disrupt the red cell button in the tube. Observe how the cells are dispersed from the red cell button. 5. Read each tube individually and record results. Use the guidelines that appear in the beginning of this manual when grading and interpreting results.
