Introductory pharmacology – Week seven learning objectives  Understand in broad terms, the major needs for antiinflammatory medication  Outline the process of inflammation and how it can change from a protective process to a damaging one.  Describe the mechanism of action of the NSAIDs, both the nonselective and the COX-2 selective.  Outline briefly how and why the non-selective COX inhibitors produce different effects from the non-selective COX inhibitors.  List the major therapeutic and adverse effects of the nonselective and selective COX inhibitors.  Outline how paracetamol differs from NSAIDs in its actions, and understand the potential of this drug to cause liver damage.  Recognise the generic names of the following drugs as nonselective NSAIDs: Aspirin, the –profens (e.g. ibuprofen, ketoprofen, carprofen)  Recognise the generic names of the following drugs as semiselective NSAIDs: the –oxicams (e.g. meloxicam, piroxicam), diclofenac.  Recognise the generic names of the following drugs as selective NSAIDs: the –coxibs (e.g.celecoxib, parecoxib)  Recognise the generic names for steroidal anti-inflammatory drugs from their –isone, -isolone, -asone endings.  Understand the mechanism of action of the steroidal antiinflammatory drugs, and their therapeutic uses.  List the major adverse effects of these drugs, and be able to relate the route of administration to the most likely side effects.  Describe how the DMARDS differ from other anti-inflammatory drugs, and how their use has to be different as a result.

 Outline the mechanism of action and adverse effects of methotrexate, leflunomide, anakinra, infliximab, abatacept and sulphasalazine  Describe how the opioids control pain, and list their major adverse effects.  Outline the mechanisms of tolerance and dependence with respect to the opioids.  Briefly describe how an opioid overdose is recognised and treated.  Apply your knowledge of the above to cases involving the therapeutic use of these drugs....
