Description

Winter 2021 Biology 2EE3

Introductory Lab 1A – Micropipetting Micropipettes are used to measure small volumes (often microlitres, µl) with a level of accuracy. The micropipettes that are used during the completion of Biology 2EE3 laboratories are models of Gilson PIPETMAN. The three models are the Gilson PIPETMAN P20, Gilson PIPETMAN P200, and Gilson PIPETMAN P1000. The parts of a Gilson PIPETMAN are not noted in the figure 1.

Figure 1. Parts of a Gilson PIPETMAN. Each of the three models has its own dispensing range as seen in table 1. The dispensing ranges is often noted on the top of the push button (sometimes also call the plunger). Table 1. Biology 2EE3 pipette and pipette ranges. Pipette

The pipettes should not be used to dispense volumes outside the of their respective specified ranges as they are not calibrated to do so. The volume that a pipette is set to dispense is noted in the volumeter display. In order to set the pipettes to a specific volume, the push button can be used as a large volume adjustment knob, and the thumbwheel can be used for fine volume adjustment. If you are using the large volume adjustment knob, or fine volume adjustment knob to decrease to a desired value/volume, decrease the value in the volumeter display and stop directly at the desired value/volume. If you are using the large volume adjustment knob, or fine volume adjustment knob to increase to a desired value/volume, increase the value in the volumeter display a few microlitres past the desired value/volume, and then slowly decrease to the desired value/volume. Examples of the volumeter displays for a P20, P200, P1000 are seen in figure 2.

Figure 2. Sample volumeter displays for Gilson PIPETMAN P20, P200, and P1000. The top value of the P20 display represents tens, the middle value ones, and the bottom value tenths of a microlitre. The top value of the P200 display represents hundreds, the middle value tens, and the bottom value represents ones in microlitres. The top value of the P1000 display represents thousands, the middle value hundreds, and the bottom values tens in microlitres. The numerical values seen in the figure 2 volumeter displays represent the following µl values that each micropipette would dispense: P20 – 14.1 µl, P200 – 141 µl, P1000 – 410 µl.
