Chromatin Immunoprecipitation (ChIP)
Approcci tradizionali al problema - Determinazione sperimentale dei siti di legame dei fattori di trascrizione. EMSA, DNAse footprinting - in vitro - Previsione computazionale dei motivi di legame. Ma un motivo può essere un vero e proprio sito di legame? - Analisi di DNA microarray di geni bersaglio. Ma il bersaglio è diretto o indiretto?
Da queste domande:
nasce la necessità di un metodo più diretto in vivo.

Chromatin immunoprecipitation (ChIP) La tecnica prevede l’identificazione di specifiche regioni/ sequenze di DNA associate con una proteina di interesse, in vivo. È diventato un potente strumento per analizzare le interazioni proteina/DNA, capace di rilevare qualsiasi segnale/modifica associato con il DNA / cromatina.
Ha avuto un enorme impatto sullo studio della cromatina, nell'epigenetica.

Reverse the crosslinking
Elution and Crosslink Reversal 1) L’aggiunta di NaHCO3/SDS causa il rilascio dell’anticorpo dalla proteina (es. Un fattore di trascrizione) (DAY 2) 2) 0.2 M NaCl a 65º per 3 h (o più) reverte il crosslink. PCR di verifica della ChIP (DAY 3) Verifica che: 1) La ChIP di controllo (PolII) è maggiore per il promotore target paragonato all’imput, il contrario si vede nel controllo negativo paragonato all’input. 2) Il controllo positivo l’Ab (polII) è maggiore rispetto al controllo negativo sul promotore target.
